Ichthyotoxic, cytotoxic and antimicrobial activity of some sponges of the Colombian Caribbean OceanDocs

The icthyotoxic, cytotoxic and antimicrobial activities of aqueous, ethanol and chloroform extractsof 11 spongesfrom the Santa Marta región, Colombian Caríbbean. were assayed. All studied sponges showed antimicrobial activity; 6 were ichthyotoxic and 7 were cytotoxic. Ircinia campana, Verongula rigida, Agelas conifera and Didiscus oxeata, were found to have activity in all three kinds of tests.

Cytotoxic and antibacterial properties of Mytilus galloprovincialis, Ostrea edulis and Crassostrea gigas (bivalve molluscs) hemolymph ArchiMer

Mussel (Mytilus galloprovincialis) plasma contains cytotoxic activity against both vertebrate (erythrocytes and mouse tumour) and protozoan cells. Procaryotes (Escherichia coli and Vibrio alginolyticus) were not sensitive to the cytotoxicity. The activity was still present in dialyzed samples but was inhibited by heating at 45°C. Large individual variability which was not correlated with protein concentration and an increasing number of reactive specimens following injection, suggested that naive mussels were in various stages of immune response. Role of nitric oxide and superoxide in Giardia lamblia killing BJMBR
Giardia lamblia trophozoites were incubated for 2 h with activated murine macrophages, nitric oxide (NO) donors or a superoxide anion generator (20 mU/ml xanthine oxidase plus 1 mM xanthine). Activated macrophages were cytotoxic to Giardia trophozoites (~60% dead trophozoites). This effect was inhibited (>90%) by an NO synthase inhibitor (200 µM) and unaffected by superoxide dismutase (SOD, 300 U/ml). Giardia trophozoites were killed by the NO donors, S-nitroso-acetyl-penicillamine (SNAP) and sodium nitroprusside (SNP) in a dose-dependent manner (LD50 300 and 50 µM, respectively). Macrophages are important components of natural immunity involved in inhibition of tumor growth and destruction of tumor cells. It is known that these cells can be activated for tumoricidal activity by lymphokines and bacterial products. We investigated whether YAC-1 tumor cells infected with Mycoplasma arginini stimulate nitric oxide (NO) release and macrophage cytotoxic activity. Thioglycollate-elicited macrophages from male BALB/c mice were co-cultured for 20 h with YAC-1 tumor cells infected or not with Mycoplasma arginini. The tumoricidal activity of activated macrophages has been attributed largely to the release of tumor necrosis factor (TNF), or to the production of reactive oxygen or nitrogen intermediates. The L929 tumor cell line (a murine fibroblast-like cell) when treated with actinomycin D (ActD) has been used to measure TNF<FONT FACE="Symbol">a</FONT> cytotoxicity. In the present study, we determined the cytotoxic activity of BCG-activated peritoneal macrophages against ActD-untreated L929 tumor cells. In vitro cytotoxicity of the LDE: daunorubicin complex in acute myelogenous leukemia blast cells BJMBR

Acute myelogenous leukemia (AML) blast cells show high-affinity degradation of low-density lipoprotein (LDL), suggesting an increased expression of cellular LDL receptors. LDE is a lipid microemulsion easily synthesized in vitro which is known to mimic the metabolic pathway of LDL. We used LDE as a carrier for daunorubicin and assayed the cytotoxicity of the complex using AML blast cells since RT-PCR analysis showed that AML cells express LDL receptor mRNA. The LDE:daunorubicin complex killed 46.7% of blast cells and 20.2% of normal bone marrow cells (P<0.001; Student t-test). Activity of the aqueous extract from Polymnia sonchifolia leaves on growth and production of aflatoxin B1 by Aspergillus flavus BJM

The aqueous extract from Polymnia sonchifolia leaves (AE) was tested for inhibitory activity on aflatoxin B1(AFB1) production and growth of Aspergillus flavus. The cytotoxicity of AE on Vero cells was also performed. Suspensions of A. flavus spores were inoculated into 50 mL of YES medium together with different concentrations of the AE. The aflatoxin B1 was extracted, analyzed by thin layer chromatography and quantified by photodensitometry. All the concentrations of AE induced inhibition of AFB1 production. The aqueous extract showed in vitro cytotoxicity to Vero cells only at concentrations above 500 µg/mL.

Reduction of adhesion properties of Ruditapes philippinarum hemocytes exposed to Vibrio tapetis ArchiMer

Vibrio tapetis is the causative agent of brown ring disease (BRD), which affects a species of clam, Ruditapes philippinarum. After incubation with V. tapetis, hemocytes lose filopods and become rounded, indicating cytotoxic activity of the bacterium. To rapidly quantify this cytotoxicity, a flow-cytometry test was developed based on the capacity of V tapetis to inhibit adhesion of clam hemocytes to plastic. Several bacteria:hemocyte ratios, the cytotoxicity of other Vibrio spp. pathogenic to bivalves, and that of various V. tapetis isolates were tested. Inhibition of adherence is detectable with as few as 5 bacteria per hemocyte. Scorpion venom toxicity is of major concern due to its influence on human activities and public health. The cytotoxicity and apoptosis induced by scorpion L. quinquestriatus venom on two established eukaryotic cell lines (293T and C2C12) were analyzed. Both cultured cell lines were incubated with varying doses (10, 20, and 50 µg/ml) of scorpion venom in serum free medium (SFM) for 0.5, 1, 2, 4, and 8 hours at 37°C. 226 methanol and water extracts representing 74 mainly native plant species found in Amazonas State, Brazil, were tested at a standard concentration of 500 μg/mL for lethality towards larvae of the brine shrimp species Artemia franciscana. Several cytotoxic plant species were identified in this work: Aspidosperma marcgravianum, A. nitidum, Croton cajucara, Citrus limetta, Geissospermum argenteum, Minquartia guianensis, Piper aduncum, P. amapense, P. capitarianum, P. tuberculatum and Protium aracouchini. The results were analyzed within the context of the available traditional knowledge and uses for these plants.

Malathion is an organophosphorate insecticide whose main acute poisonous effect is the inhibition of the enzyme acetylcholinesterase. This pesticide is widely used and has been reported to damage mice spermatogenesis. The objective of this work was to evaluate the toxic risk of malathion, earthworm Eisenia foetida (Annelida, Oligochaeta) like biological model was used and testicles and spermatozoa of animal controls and treaties were analyzed. Earthworms with developed clitellum were used and they were dewed with sublethal single doses of 0, 80, 150, 300 and 600mg of malathion for kilogram of soil (LD50 = 880mg/kg dry soil). We investigated the in vitro process of cell death caused by Egyptian cobra venom on primary human embryonic kidney (293T) and mouse myoblast (C2C12) cell lines. The aim of these studies was to provide further information about triggering cell death, and suggest methods for eliminating unwanted cells, such as tumour cells. Chlorhexidine, even at low concentrations, is toxic for a variety of eukaryotic cells; however, its effects on host immune cells are not well known. We evaluated in vitro chlorhexidine-induced cytotoxicity and its effects on reactive oxygen/nitrogen intermediate induction by murine peritoneal macrophages. Thioglycollate-induced cells were obtained from Swiss mice by peritoneal lavage with 5 ml of 10 mM phosphate-buffered saline, washed twice and resuspended (10(6) cells/ml) in appropriate medium for each test. Dipyridamole increases the cytotoxicity of cisplatin in human larynx cancer cells in vitro BJMBR

This paper describes the effect of dipyridamole (DIP) on the cytotoxicity of cisplatin in HEp-2 human larynx cancer cells in vitro and the nature of the interaction between cisplatin and dipyridamole. Cytotoxic assays were performed to obtain the IC50 for cisplatin. The cells were treated with 0, 20, 40, 80, 120 or 200 µM cisplatin, with or without a single concentration of DIP and incubated for 60 min at 37ºC and 5% CO2 for 3 days and then counted with a hemocytometer. The accumulation of cisplatin in the cells was measured by atomic absorption and fluorescence was used to determine the membrane binding constant of DIP. Mercury is a xenobiotic metal that is a highly deleterious environmental pollutant. The biotransformation of mercury chloride (HgCl2) into methylmercury chloride (CH3HgCl) in aquatic environments is well-known and humans are exposed by consumption of contaminated fish, shellfish and algae. The objective of the present study was to determine the changes induced in vitro by two mercury compounds (HgCl2 and CH3HgCl) in cultured human lymphocytes. Cytotoxicity of subfractions and compounds from Polymnia sonchifolia BJM

Our previous works showed that extracts and fractions from Polymnia sonchifolia had presented an inhibitory activity on growth and aflatoxin production by Aspergillus flavus in non-cytotoxic concentrations. In this work, report results on the in vitro cytotoxicity of subfractions and of a mixture of sesquiterpenes lactones from Polymnia sonchifolia on Vero cells. The cytotoxicity was assayed through crystal violet staining (CVS) method and the 50% inhibitory concentration for cell growth (IC50) was calculated. Both subfractions and the mixture of sesquiterpenes lactones did not show in vitro cytotoxicity to Vero cells on active biologically concentrations against production of aflatoxin B1 by Aspergillus flavus.

This study evaluates the biological activity and possible deterrance played by organic crude extracts from six Phaeophyta and two Rhodophyta algae from Southern Caribbean. Bioessays were carried out using concentrations equivalent to those present in algae tissue. Species in general did not show high citotoxic activity. However Dictyota pulchella, showed a delay in the mitotic process for cells stage XVI. Extracts from Dictyota bartayresiana, D. pulchella, Sargassum cymosum and Cladophyllum schnetteri, inhibited S. partitus feeding under laboratory conditions. Nontheless, only extracts from D. bartayresiana and C. schnetteri showed this activity in the field.

In this study, the onion (Allium cepa) root test was used to evaluate the genotoxicity of the Pitimbu River (Natal city, Brazil) surface water. The water was collected at five sampling sites along the river and one sample was obtained after the treatment (flocculation, chlorination and pH correction) of the river water for human consumption. All raw river water samples increased the frequency of chromosomal abnormalities and/or micronuclei and two of the water samples produced alterations in the mitotic index of the root cells. Two of the water samples also altered root growth and two produced morphological modifications in the A. cepa roots. Water collected from a site near an industrial area was the most consistently toxic and genotoxic of the samples....

La determinación de actividad antimitótica de extractos de organismos marinos genera expectativas para la obtención de sustancias con potencial anticancerígeno a partir de fuentes alternativas. Se evaluó la actividad antimitótica in vitro de extractos orgánicos crudos de 15 esponjas de la costa Caribe colombiana, en embriones del erizo de mar Lytechinus variegatus.
